Abstract

Pemajanan akut dan subkronik MP-PE dalam dosis rendah hingga tinggi dapat menyebabkan penurunan fungsi ginjal dan hati dengan tanda peningkatan kadar kreatinin, SGOT dan SGPT, kerusakan organ hati dan ginjal pada tikus wistar bahkan risiko kesehatan jangka panjang bagi manusia yang mengkonsumsi air minum yang terkontaminasi MP-PE, tujuan dalam penelitian ini adalah (a) menganalisis efek akut dari pajanan MP-PE yang dinyatakan dalam nilai LD50 terhadap tikus wistar (rattus norvegicus); (b) menganalisis peningkatan kadar kreatinin ginjal serta SGOT dan SGPT pada pajanan akut dan subkronik MP-PE terhadap tikus wistar (rattus norvegicus); (c) menganalisis gambaran histopatologis hati dan ginjal akibat pajanan akut dan subkronik MP-PE terhadap tikus wistar (rattus norvegicus), (d) menganalisis risiko kesehatan akibat pajanan MP-PE dengan metode ARKL (analisis risiko kesehatan lingkungan). Desain penelitian adalah eksperimental laboratorik pada tikus tikus wistar (rattus norvegicus) berjenis kelamin jantan dengan menggunakan desain penelitian pre and post test control group design, dilanjutkan dengan terminate hewan uji untuk pemeriksaan gambaran histapotalogis organ ginjal dan hati, kemudian identifikasi bahaya MP-PE bagi kesehatan dilakukan dengan melakukan pengambilan sampel lingkungan yang mengandung MP-PE yaitu air minum masyarakat yang diperoleh melalui air sumur gali, analisis dosis respon dengan dengan cara melihat alur pajanan dalam lingkungan dengan menghitung berat badan masyarakat, lama konsumsi, dan lowest observable adverse effect Level (LOAEL). Berdasarkan hasil penelitian bahwa MP-PE memiliki dampak yang signifikan terhadap perubahan berat badan, peningkatan kadar kreatinin, SGOT, dan SGPT tikus serta kelainan histopatologis, pada pajanan akut, adanya 1 kematian, menunjukkan bahwa tidak didapatkannya f yaitu yang diperoleh dari tabel Thomson dan Weil, sehingga nilai LD50 tidak dapat dihitung dan dapat disimpulkan bahwa MP-PE tidak menimbulkan kematian 50%, namun untuk kategori keracunan maka kematian satu ekor tikus dapat dismpulkan bahwa bahan MP-PE dapat memberikan efek dengan kategori satu (toksisitas ringan) karena dapat dibuktikan dengan hasil histopatologis tikus yang telah mulai mengalami kerusakan pada dosis rendah 5 mg/kgBB, pada pajanan akut dan subkronis ditemukan peningkatan kadar kreatinin, SGOT, dan SGPT juga menunjukan kerusakan organ ginjal dan hati. Kelimpahan partikel MP-PE pada air minum masyarakat terdeteksi maksimum 269 partikel/m3 sehingga MP-PE berisiko terhadap kesehatan masyarakat karena air minum tersebut dikonsumsi oleh masyarakat. Kesimpulan dalam penelitian ini yaitu MP-PE secara akut dan subkronis menimbulkan pengaruh terhadap penurunan fungsi hati dan ginjal tikus pre dan post dari dosis MP-PE dan juga menyebabkan kerusakan organ hati dan ginjal dimana ginjal mengalami peradangan dengan status sweeling hingga hyalin cast dan hati mengalami peradangan inflammation hingga steatosis. Lebih burukunya pajanan MP-PE terdeteksi banyak pada sumber air minum masyarakat dan setelah dihitung dengan menggunakan metode analisis risiko Kesehatan lingkungan (ARKL) MP-PE berakibat pada risiko kesehatan non-karsinogenik baik bagi anak-anak maupun orang dewasa melalui jalur konsumsi dengan nilai RQ > 1.

Acute and subchronic exposure to MP-PE in low to high doses can cause a decrease in kidney and liver function with signs of increased levels of creatinine, SGOT and SGPT, liver and kidney damage in Wistar rats and even long-term health risks for humans who consume contaminated drinking water. MP-PE, the objectives of this study were (a) to analyze the acute effects of MP-PE exposure expressed in LD50 values on Wistar rats (rattus norvegicus); (b) analyzing the increase in renal creatinine levels as well as SGOT and SGPT on acute and subchronic exposure to MP-PE on Wistar rats (rattus norvegicus); (c) analyzing the histopathological features of the liver and kidneys due to acute and subchronic MP-PE exposure to Wistar rats (rattus norvegicus), (d) analyzing the health risks due to exposure to MP-PE using the ARKL method (environmental health risk analysis). The research design was laboratory experimental on male Wistar rats (rattus norvegicus) using a pre and post test control group design, followed by terminating the test animals to examine the histapotological features of the kidneys and liver, then identifying the dangers of MP-PE for health carried out by taking environmental samples containing MP-PE, namely community drinking water obtained through dug well water, dose response analysis by looking at the flow of exposure in the environment by calculating people's body weight, duration of consumption, and the lowest observable adverse effect level (LOAEL ). Based on the research results that MP-PE has a significant impact on changes in body weight, increased levels of creatinine, SGOT, and SGPT in mice as well as histopathological abnormalities, in acute exposure, there was 1 death, indicating that f was not obtained, namely that obtained from the Thomson and Weil tables. , so the LD50 value cannot be calculated and it can be concluded that MP-PE does not cause 50% mortality, but for the poisoning category, the death of one rat can be concluded that the MP-PE material can have an effect in category one (mild toxicity) because it can be proven by histopathological results of mice that had begun to experience damage at a low dose of 5 mg/kgBW, in acute and subchronic exposure found increased levels of creatinine, SGOT, and SGPT also showing damage to the kidneys and liver. The maximum abundance of MP-PE particles in community drinking water was detected at 269 particles/m3, making MP-PE a risk to public health because drinking water is consumed by the community. The conclusion in this study is that MP-PE acutely and subchronically has an effect on reducing the liver and kidney function of mice pre and post dose of MP-PE and also causes damage to the liver and kidneys where the kidneys experience inflammation with a sweeling status to hyaline casts and the liver experiences inflammation to steatosis. Worse, MP-PE exposure was detected in many community drinking water sources and after being calculated using the Environmental Health Risk Analysis (ARKL) method, MP-PE resulted in non-carcinogenic health risks for both children and adults through the consumption route with an RQ value. > 1.
