Abstract

epidermidis. Wilis Ari Setyati & Sri Sedjati).
Kesehatan merupakan hal yang esensial dalam kehidupan setiap individu.
Penyakit kulit merupakan penyakit non-fatal yang dapat mempengaruhi 30-70%
individu secara global tanpa memandang kelompok umur. Penyakit kulit pada
manusia disebabkan oleh infeksi patogen seperti bakteri Staphylococcus sp.,
Streptococcus sp., dan jamur seperti Candida sp. dan Malassezia. Penyakit kulit
umumnya ditangani dengan pemberian antibiotik. Pemberian antibiotik menjadi
tidak efektif lagi akibat resistensi dari patogen penyebab infeksi kulit. Oleh karena
itu, penemuan antibiotik baru untuk mengatasi patogen penyebab penyakit kulit
penting dilakukan. Salah satu sumber bahan hayati laut yang menarik untuk diteliti
adalah kapang endofit mangrove. Kapang endofit mangrove diketahui memiliki
senyawa bioaktif yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk
mengisolasi kapang endofit mangrove Rhizophora apiculata yang memiliki
kemampuan melawan bakteri Staphylococcus epidermidis. Isolat kapang yang
diketahui memiliki aktivitas antibakteri kemudian diuji untuk menentukan
konsentrasi ekstrak kapang yang diperlukan untuk menghambat dan membunuh
pertumbuhan bakteri tersebut, serta mengidentifikasi jenis senyawa metabolit
sekunder yang terkandung di dalam ekstrak kapang. Metode yang digunakan dalam
penelitian ini meliputi pengambilan sampel, isolasi dan purifikasi kapang, skrining
aktivitas antibakteri metode agar plug, identifikasi makroskopis, identifikasi
molekuler dengan ekstraksi DNA menggunakan metode Chelex, uji MIC dengan
metode mikrodilusi, uji MBC, serta uji TLC. Hasil penelitian ini menunjukkan
bahwa terdapat satu spesies kapang endofit mangrove Rhizophora apiculata dengan
diberi kode MG.05.T.15 yang memiliki aktivitas antibakteri melawan bakteri
Staphylococcus epidermidis dengan konsentrasi minimum sebesar 2,5 mg/mL
untuk menghambat pertumbuhan bakteri patogen dan konsentrasi sebesar 5 mg/mL
untuk membunuh pertumbuhan bakteri patogen tersebut. Hasil identifikasi
molekuler menunjukkan bahwa isolat MG.05.T.15 memiliki kemiripan dengan
Diaporthe sennae dengan homologi sebesar 96,52%. Ekstrak etil asetat isolat
potensial diketahui mengandung senyawa metabolit sekunder yaitu fenol,
flavonoid, dan terpenoid. Ekstrak metanol isolat potensial mengandung senyawa
metabolit sekunder yaitu alkaloid, flavonoid, dan terpenoid.

Health is an essential part of every individual's life. Skin health is important
to maintain in order to prevent skin disease infections. Skin diseases in humans are
caused by pathogenic infections of bacteria such as Staphylococcus sp.,
Streptococcus sp., and fungi such as Candida sp. and Malassezia. Skin diseases are
usually treated with antibiotics. Antibiotic treatment is no longer effective anymore
due to resistance from pathogens that cause skin infections. Therefore, the
discovery of new antibiotics to against pathogen that cause skin diseases is
important. One of the interesting sources of marine biological resources to be
studied is mangrove endophytic fungi. Mangrove endophytic fungi are known have
bioactive compounds that potentially as antibacterial.This research aims to isolate
the mangrove endophytic fungi from Rhizophora apiculata that have antibacterial
acitivity against Staphylococcus epidermidis bacteria. The isolate that known
having antibacterial acitivity then tested to determine the concentration of fungi
extract needed to inhibit and kill the growth of that bacteria, as well as to identify
the types of secondary metabolites compounds that contained in the extract. The
methods used in this research included sampling, isolation and purification of fungi,
screening for antibacterial activity using the agar plug method, macroscopic
identification, molecular identification by DNA extraction using the Chelex method,
MIC assay using the microdilution method, MBC assay, and TLC assay. The results
of this research indicate that there is one species of mangrove endophytic fungi
from Rhizophora apiculata coded MG.05.T.15 which has antibacterial activity
against Staphylococcus epidermidis bacteria with a minimum concentration of 2,5
mg/mL to inhibit the growth of pathogenic bacteria and a concentration of 5
mg/mL to kill the growth of this pathogenic bacteria. The result of molecular
identification showed that strain isolate MG.05.T.15 has similarity with Diaporthe
sennae with homology 96.52%. Ethyl acetate extract of potential isolates is known
containing secondary metabolite compounds such as phenols, flavonoids, and
terpenoids. Methanol extract of potential isolates contains secondary metabolite
compounds such as alkaloids, flavonoids, and terpenoids.
