Abstract

(Febriansyah Hanik Dwi Saputri. 26060119130074. Optimasi Suhu dan Waktu Ekstraksi Pigmen Thalassiosira sp. dengan Minyak VCO Menggunakan Response Surface Methodology Eko Susanto dan Putut Har Riyadi).
Thalassiosira sp. merupakan tumbuhan mikroalga yang kaya akan pigmen klorofil, karotenoid dan bioaktif lainnya seperti polisakarida, asam lemak tak jenuh. Kandungan yang terdapat pada Thalassiosira sp. terutama pigmen berpotensi besar untuk kesehatan manusia, hal ini mampu menjadikan Thalassiosira sp. sebagai bahan tambahan pangan fungsional. Ekstraksi pigmen dengan VCO dan mikroalga dapat langsung diektraksi dengan metode maserasi dan ekstraksi hijau. Hasil ekstrak diharapkan menunjukkan aktivitas biologis gabungan yang berasal dari fitokimia seperti pigmen. Penelitian ini bertujuan untuk mengkaji pengaruh suhu ekstraksi dan waktu ekstraksi terhadap kualitas pigmen VCO yang diperkaya ekstrak Thalassiosira sp. serta menentukan suhu ekstraksi dan waktu ekstraksi optimum menggunakan metode Response Surface Methodology. Suhu ekstraksi yang digunakan yaitu 50 s.d. 70℃ sedangkan waktu ekstraksi yang digunakan yaitu 60 s.d. 120 menit. Data uji kadar pigmen dan antioksidan dianalisis dengan metode RSM menggunakan Design Expert 13. Hasil penelitian menunjukkan bahwa suhu ekstraksi dan waktu ekstraksi berpengaruh nyata terhadap hasil nilai kadar pigmen dan antioksidan. Hasil solusi formula optimasi diperoleh suhu ekstraksi 60℃ dan waktu ekstraksi 90 menit yang menghasilkan VCO yang diperkaya ekstrak
Thalassiosira sp. dengan respon optimum yaitu pigmen klorofil a sebesar 13,5723 µg/mL, pigmen klorofil b 10,6861 µg/mL, pigmen total klorofil 24,248 µg/mL dan pigmen karotenoid sebesar 9,7407 µg/mL. Perlakuan optimum menghasilkan pigmen pheophytin a sebesar 23,3289 µg/mL, pigmen pheophytin b sebesar 87,534 µg/mL, pigmen total pheophytin sebesar 43,7173 µg/mL dan antioksidan sebesar 11,1214% dengan nilai desirability 0,917.

Temperature and Time of Pigment Extraction of Thalassiosira sp. with VCO Oil Using Response Surface Methodology Eko Susanto and Putut Har Riyadi).
Thalassiosira sp. is a microalgae plant that is rich in chlorophyll,
carotenoid and other bioactive pigments such as polysaccharides, unsaturated fatty acids. The content contained in Thalassiosira sp. especially pigments have great potential for human health, this can make Thalassiosira sp. as a functional food additive. Pigment extraction with VCO and microalgae can be directly extracted
by maceration and green extraction methods. The resulting extract is expected to exhibit combined biological activity derived from phytochemicals such as pigments. This study aims to examine the effect of extraction temperature and extraction time on the quality of VCO pigments enriched with Thalassiosira sp. extract. and
determine the optimum extraction temperature and extraction time using Response Surface Methodology method. The extraction temperature used was 50 s.d. 70 ℃ while the extraction time used is 60 s.d. 120 minutes. Pigment and antioxidant test data were analyzed using the RSM method using Design Expert 13. The results showed that the extraction temperature and extraction time had a significant effect on the pigment and antioxidant levels. The results of the optimization formula solution obtained an extraction temperature of 60 ℃ and an extraction time of 90
minutes which produced VCO enriched with Thalassiosira sp. with the optimum response of chlorophyll a pigment of 13.5723 µg/mL, chlorophyll b pigment of 10.6861 µg/mL, total chlorophyll pigment of 24.248 µg/mL and carotenoid pigment of 9.7407 µg/mL. The optimum treatment produced pheophytin a pigment of 23.3289 µg/mL, pheophytin b pigment of 87.534 µg/mL, total pheophytin pigment of 43.7173 µg/mL and antioxidant of 11.1214% with a desirability value of 0.917.
