Abstract

ABSTRAK

Pengkayaan Asam Askorbat dan Instan Yeast Terhadap Uji Kejut Salinitas.
Subagiyo dan Ervia Yudiati).
Artemia sp. termasuk dalam pakan alami yang digunakan untuk budidaya.
Kandungan gizi utama pada Artemia sp. yaitu protein dan asam amino.
Kandungan gizi yang ada pada Artemia sp masih belum optimal untuk
meningkatkan kelangsungan hidunya, maka dari itu diperlukannya pengkayaan
pada Artemia sp. Tujuan dari penelitian ini yaitu mengetahui tingkat toksisitas
asam askorbat sebagai pakan Artemia sp. dan pengaruh pengkayaan asam askorbat
dan fermipan pada pakan terhadap uji kejut salinitas Artemia sp. Metode
penelitian yang digunakan yaitu metode eksperimental laboratoris dengan
Rancangan Acak Lengkap (RAL). Uji toksisitas Artemia sp. pada asam askorbat
menggunakan metode Brine Shirimp Lethality Test (BSLT). Nilai toksisitas
digunakan dalam menentukan dosis perlakuan. Pengkayaan Artemia sp. dilakukan
dengan cara perendaman selama 3 hari yang terdiri atas 8 perlakuan dan kontrol.
Perlakuan tersebut yaitu FR (Fermipan 0,15 g/500mL), AS 1 (asam askorbat 0,05
g/500mL), AS 2 (asam askorbat 0,025 g/500mL), AS 3 (asam askorbat 0,0125
g/500mL), C1 (asam askorbat 0,025 g/250mL dan fermipan 0,0075 g/250mL), C2
(asam askorbat 0,0125 g/250mL dan fermipan 0,0075 g/250mL), C3 (asam
askorbat 0,00625 g/250mL dan fermipan 0,0075 g/250mL) dengan masingmasing 3 pengulangan. Uji kejut salinitas dilakukan dengan menempatkan
Artemia sp. dari salinitas 25 ppt ke 0 ppt. Tingkat kelangsungan hidup dicatat
setiap 4 jam sekali hingga Artemia sp. mati. Hasil penelitian uji toksisitas dengan
nilai LC50 yaitu 200,84 < 1000 ppm, yang menunjukkan sifat toksik asam askorbat
terhadap Artemia sp. Ketahanan uji salinitas pada Artemia sp. dengan kurun waktu
terlama secara berurutan terdapat pada perlakuan AS 2 (20 jam); FR, C1, dan C2
(16 jam); AS 3, C3 dan kontol (12 jam) dan AS 1 (8 jam). Pengkayaan asam
askorbat dan fermipan dalam pakan Artemia sp. berpengaruh nyata terhadap uji
kejut salinitas. Kombinasi asam askorbat dan fermipan tidak berpengaruh nyata
terhadap uji kejut salinitas Artemia sp.

Artemia sp. included in the natural feed used for cultivation. The main
nutrional content in Artemia sp. namely proteins and amino acids. The nutritional
content in Artemia sp. is stil not optimal to increase its maintenance of life,
therefore it it necessary to enrich Artemia sp. and the effect of enrichment of
ascorbid acid and fermipant in feed on the salinity shock test of Artemia sp. The
research method used was a laboratory experimental method with a completely
randomized design (CRD). Artemia sp toxicity test to ascorbid acid using the
Brine Shririmp Lethality Test (BSLT) method. Toxicity value used in determining
the dose of treatment. Enrichment of Artemia sp. done by soaking for 3 days
consisting of 8 treatments were FR (Fermian 0,15 g/50 mL), AS 1 (ascorbid acid
0,05 g/500 ml), AS 2 (ascorbid acid 0,025 g/500 mL), AS 3 (ascorbid acid 0,0125
g/500 mL), C1 (ascorbid acid 0,025 g/250 mL and fermipan 0,0075 g/250 mL),
C2 (ascorbid acid 0,0125 g/250 mL and fermipan 0,0075 g/250 mL), C3 (ascorbid
acid 0,00625 g/250 mL and fermipan 0,0075 g/250 mL) with repetitions each.
Salinity test was carried out by placing Artemia sp. cheese from 25 ppt to 0 ppt
salinity. The level of regret for life was recorded every 4 hours until Artemia sp.
dead. The results of toxicity test study with an LC50 value of 200,84 < 1000 ppm,
which indicates the toxic properties of ascorbid acid against Artemia sp. Salinity
test resistance on Artemia sp. with the longest time limit consecutively in the AS 2
treatment (20 hours); FR, C1 and C2 (16 hours); AS 3, C3and dick (12 hours)
and AS 1 (8 hours). Enrichment of ascorbid acid and fermipan in feed Artemia sp.
significant effect on the salinity shock test. The combination of ascorbid acid and
fermipan did not significantly affect the salinity shock test of Artemia sp.
