Abstract

(Alvinola Prisca Fatina. 26040119130080. Potensi Abalon Haliotis asinina Sebagai Antibakteri dan Pengaruh Terhadap Kadar Kolesterol dan Glukosa Darah Mencit Mus musculus. Delianis Pringgenies dan Ali Ridlo).
Abalon spesies Haliotis asinina memiliki banyak manfaat karena mengandung nutrisi dan senyawa bioaktif yang dapat dimanfaatkan dalam bidang bioteknologi dan farmakologi laut seperti sebagai antibakteri dan pengaruhnya terhadap kolesterol dan glukosa darah. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak abalone terhadap bakteri S. aureus dan E.coli serta untuk mengetahui pengaruhnya pada kolesterol dan glukosa darah mencit Mus musculus. Ekstraksi sampel abalone H. asinina dilakukan dengan metode maserasi bertingkat menggunakan pelarut n-heksana, etil asetat, dan metanol. Uji aktivitas antibakteri dilakukan secara kualitatif dengan metode difusi cakram menggunakan media uji MHA (Mueller Hinton Agar) dan pengamatan zona bening yang dilakukan selama 3x24 jam. Uji kadar kolesterol dan glukosa darah mencit dilakukan menggunakan 5 ekor mencit yang terbagi atas 5 kelompok dosis yaitu kontrol positif, kontrol negatif, dosis ekstrak metanol abalone 100, 200, dan 300 mg/kgBB yang diamati setiap 14 hari selama 28 hari. Hasil penelitian menunjukkan bahwa terdapat aktivitas antibakteri ekstrak metanol pada konsentrasi 20% terhadap bakteri S. aureus, ekstrak etil asetat abalone H. asinina pada konsentrasi 10% dan 20% menunjukkan adanya zona hambat terhadap bakteri S. aureus dan E. coli sedangkan pada ekstrak n-heksana terdapat aktivitas antibakteri pada konsentrasi 10% terhadap bakteri uji S. aureus dan E.coli dan konsentrasi 20% terhadap bakteri uji S. aureus. Ekstrak metanol daging abalone H. asinina menunjukkan dapat menurunkan kolesterol pada dosis 300 mg/kg sedangkan pada uji glukosa darah tidak menunjukkan aktivitas penurunan pada semua dosis uji. Hasil uji fitokimia pada ekstrak metanol abalone H. asinina mengandung saponin dan alkaloid.

Abalone species Haliotis asinina has many benefits by containing nutrients and bioactive compounds that can be utilized in the field of marine biotechnology and pharmacology such as antibacterial and its effect on cholesterol and blood glucose. This study aims to determine the antibacterial activity of abalone extract against S. aureus and E. coli bacteria and to determine its effect on cholesterol and blood glucose of Mus musculus mice. The extraction of H. asinina abalone samples was conducted by graded maceration method using methanol, ethyl acetate, and nhexane solvents. The antibacterial activity test was carried out qualitatively using the disc diffusion method using MHA (Mueller Hinton Agar) test media and observation of clear zones carried out for 3x24 hours. The test of cholesterol and blood glucose levels in mice was carried out using 5 mice which were divided into 5 dose groups, which are positive control, negative control, doses of abalone methanol extract 100, 200, and 300 mg/kgBB which were observed every 14 days for 28 days. The results showed that there was antibacterial activity of methanol extract with a concentration of 20% on S. aureus bacteria, in the ethyl acetate extract of abalone H. asinina at a concentration of 10% and 20% showed an inhibition zone against S. aureus and E. coli bacteria while in the n-hexane extract there was antibacterial with a concentration of 10% on the test bacteria S. aureus and E.coli and a concentration of 20% on the test bacteria S. aureus. The methanol extract of H. asinina abalone shows that it can reduce cholesterol at a dose of 300 mg/kg while the blood glucose test does not show a decrease in activity at various test doses. The results of phytochemical tests on methanol extract of abalone H. asinina contain saponins and alkaloids.
