(Afina Dwyna Inayah. 26040119130066. Pertumbuhan dan Kandungan Karotenoid Porphyridium cruentum pada Intensitas Cahaya Berbeda Skala Laboratorium. Gunawan Widi Santosa dan Widianingsih).

Porphyridium cruentum merupakan salah satu mikroalga dari kelompok alga merah (Rhodophyta). P. cruentum mengandung beberapa senyawa bioaktif salah satunya karotenoid. Karotenoid merupakan salah satu kelompok antioksidan alami yang dapat digunakan sebag ai sumber antioksidan tambahan melalui pangan fungsional. Salah satu faktor yang mempengaruhi produksi karotenoid dalam mikroalga adalah intensitas cahaya. Penelitian ini bertujuan untuk mengetahui bagaimana pengaruh perbedaan intensitas cahaya terhadap laju pertumbuhan serta kandungan karotenoid dalam mikroalga P. cruentum. Metode yang digunakan adalah eksperimen laboratorium dengan menggunakan Rancangan Acak Lengkap (RAL). Mikroalga P. cruentum dikultivasi dengan tiga taraf perlakuan intensitas cahaya yang berbeda yaitu 2000 lux, 4000 lux, 5000 lux dan 3000 lux (kontrol) dengan masing-masing tiga pengulangan. Pertumbuhan P.cruentum diamati selama 8x24 jam dan kemudian dipanen untuk perhitungan biomassanya. Biomassa basah hasil kultivasi diekstraksi dengan pelarut aseton PA. Ekstrak P. cruentum kemudian dianalisis kandungan karotenoidnya secara spektrofotometri. Laju pertumbuhan P. cruentum tertinggi didapatkan pada intensitas cahaya 5000 lux dengan nilai 0,229 sel/mL/hari. Kandungan karotenoid P. cruentum tertinggi didapatkan pada perlakuan intensitas cahaya 4000 lux dengan nilai 13,530 µg/mL. Berdasarkan hasil penelitian dapat disimpulkan bahwa perlakuan intensitas cahaya yang berbeda berpengaruh secara signifikan terhadap laju pertumbuhan P. cruentum, namun tidak berpengaruh secara signifikan terhadap kandungan karotenoid pada P. cruentum.

Porphyridium cruentum is one of the microalge from the red algae group (Rhodophyta). P. cruentum contains several bioactive compounds, such as carotenoid. Carotenoids are considered as a natural antioxidants that can be used as additional sources of antioxidants through functional foods. One of the factors that affecting carotenoid production in microalgae is light intensity. The aim of this study is to determine the effect of varying light intensity on growth rate and carotenoid content in P. cruentum microalgae. The method used in this study is an experimental laboratory which analyzes by a Completely Randomized Design (CRD). P. cruentum microalgae were cultivated with three varieties of light intensity treatment, which are 2000 lux, 4000 lux, 5000 lux and 3000 lux (control) with three replications each. The growth of P. cruentum was observed for 8x24 hours then was harvested to calculate its biomass. The wet cultivated biomass was extracted with PA acetone solvent. The P. cruentum extract was analyzed for its carotenoid content by spectrophotometry. The highest growth rate was found in 5000 lux of light intensity with total 0,229/cell/mL/day. The highest carotenoid content was found in 4000 lux of light intensity with total 13.530 µg/mL. It can be concluded that varying light intensity has a significant effect on the growth rate, however not on the carotenoid content in P. cruentum.
