Abstract

ABSTRAK
(Fahma Gumilang Hendrasjati. 26020117130076. Respon Pemberian Ekstrak Tembakau (Nicotiana tobacum) yang Berbeda sebagai Agen Anestesi terhadap Kelulushidupan pada Transportasi Benih Ikan Koi (Cyprinus carpio Koi). Istiyanto Samidjan dan Desrina) Ikan Koi (Cyprinus carpio Koi) merupakan komoditas budidaya yang mempunyai nilai ekonomi tinggi bagi masyarakat. Salah satu tahapan budidaya ikan koi adalah pendistribusian ikan kepada konsumen. Tahapan ini memiliki peranan penting karena memiliki resiko kematian yang tinggi dikarenakan ikan mengalami stres. Salah satu upaya yang dapat dilakukan untuk mengantisipasi ikan stres adalah dengan menekan aktivitas metabolisme ikan Bahan yang sering digunakan pada anestesi yaitu tricaine methanesulphonate atau yang lebih dikenal dengan MS-222 bahan sintetik yang memiliki harga yang sangat mahal Salah satu alternatif yang dapat digunakan yaitu dengan menggunakan senyawa yang terkandung dalam ekstrak tembakau (Nicotiana tobaccum). Tembakau memiliki kandungan senyawa flavonoid, saponin, dan nikotin. Tujuan dari penelitian ini adalah untuk mengkaji pengaruh perbedaan pemberian ekstrak tembakau dengan konsentrasi yang berbeda dalam sistem transportasi tertutup. Metode penelitian yang digunakan adalah metode eksperimental menggunakan Rancangan Acak Lengkap dengan 5 perlakuan dan 3 ulangan, dengan perlakuan A (0 ml/l), B (3,15 ml/l), C (6,3 ml/l), D (9,45ml/l), E (12,6 ml/l). Ikan koi yang digunakan berukuran 5cm, diangkut dengan sistem tranportasi tertutup menggunakan plastik polyethylene diisi dengan 10 benih ikan koi dan diisi air sebanyak 6 liter perkantong plastik.
Hasil penelitian menunjukkan bahwa perbedaan pemberian jumlah konsetrasi ekstrak tembakau (Nicotiana tobacum) berpengaruh sangat nyata dalam laju pemingsanan dan pemulihan, kelulushidupan dan glukosa darah (P<0,01). Perubahan tingkah laku diamati dengan melihat bukaan operculum, respon terhadap rangsangan luar, dan keseimbangan ikan. Laju waktu pemingsanan berbanding terbalik terhadap konsentrasi ekstrak, sedangkan laju pemulihan berbanding lurus terhadap konsentrasi ektrak. Namun tingkat kelulushidupan pada setiap perlakuan yaitu B (3,15 ml/l), C (6,3 ml/l), D (9,45ml/l), E (12,6 ml/l) kecuali A (0 ml/l) sebagai kontrol memiliki tingkat kelulushidupan dibawah 80%. Glukosa dalam darah mengalami penuruna signifikan pada perlakuan B dan C yaitu 89,33 mg/dl dan 106,00 mg/dl.

(Fahma Gumilang Hendrasjati. 26020117130076. Response of Different Tobacco Extract (Nicotiana tobacum) as Anesthesia Agent on Survival in Transportation of Koi Fish (Cyprinus carpio Koi). Istiyanto Samidjan and Desrina) Koi fish (Cyprinus carpio Koi) is a cultivated commodity that has high economic value for the community. One of the stages of koi fish cultivation is the distribution of fish to consumers. This stage has an important role because it has a high risk of death. One of the efforts that can be made to anticipate stress in fish is to suppress fish metabolic activity. Materials that are often used in anesthesia are synthetic materials which are very expensive. One of them is tricaine methanesulphonate or better known as MS-222. One alternative that can be used is to use compounds contained in tobacco extract (Nicotiana tobaccum). Tobacco contains flavonoids, saponins and nicotine. The purpose of this study was to examine the effect of different concentrations of tobacco extract in a closed transport system. The research method used was an experimental method using a completely randomized design with 5 treatments and 3 replications. The treatment used was using differen tobacco extracts (Nicotiana tobaccum), namely treatment A (0 ml/l), B (3.15 ml/l), C (6.3 ml/l), D (9.45 ml/l ), E (12.6 ml/l). The koi fish used are 5cm sized seeds, transported in a closed transportation system using polyethylene plastic filled with 10 koi fish seeds and filled with 6 liters of water per plastic bag. The results showed that the difference in the amount of concentration of tobacco extract (Nicotiana tobacum) had a very significant effect. in stunning and
recovery rates, survival and blood glucose (P<0.01). Changes in behavior were observed by looking at the opening of the operculum, the response to external stimuli, and the balance of the fish. The stunning time rate is directly proportional to the extract concentration, while the recovery rate is inversely proportional to the extract concentration. However, the survival rates for each treatment were B (3.15 ml/l), C (6.3 ml/l), D (9.45 ml/l), E (12.6 ml/l) except A (0 ml /l) as a control had a survival rate below 80%. Glucose in the blood decreased significantly in treatment B and C, namely 89.33 mg/dl and 106.00 mg/dl.
