Bakteri Rhizosfer dari Ekosistem Mangrove di Tapak, Kecamatan Tugu, Kota
Semarang (Aninditia Sabdaningsih dan Oktavianto Eko Jati)
Ekosistem mangrove di Tapak merupakan ekosistem terletak di sepanjang Sungai
Tapak, Semarang dan dikelilingi oleh tambak tradisional ikan bandeng dan udang
vaname. Sungai Tapak merupakan tempat pembuangan limbah industri sabun.
Letak dan kondisi tersebut menjadikannya kaya akan unsur hara yang terkandung
dalam sedimen khususnya rhizosfer atau area sedimen yang berada di sekitar
perakaran. Hal tersebut mendukung berbagai kehidupan mikroba di dalamnya
yang berperan dalam proses siklus nutrien. Tujuan dari penelitian ini yaitu untuk
mengisolasi bakteri, mengetahui kelimpahan bakteri serta mengidentifikasi jenis
bakteri rhizosfer yang kemudian diujikan kemampuannya sebagai pelarut fosfat.
Penelitian ini menggunakan pendekatan eksploratif. Waktu pengambilan sampel
dilaksanakan pada bulan November 2021 dan analisis sampel dilakukan pada
bulan November 2021 – Juni 2022. Sampel sedimen diambil pada 4 stasiun
menggunakan sediment core. Sampel sedimen kemudian diisolasi pada media
GSP (Glutamate Starch Agar) dan nutrient agar untuk mendapatkan isolat murni.
Sebanyak 22 isolat diperoleh dengan 11 isolat Gram positif dan 11 isolat Gram
negatif, seluruh bentuk sel yang didapatkan adalah basil. Kelimpahan bakteri
tertinggi didapatkan pada stasiun A sebesar 1,44 × 104 CFU/mL dan kelimpahan
terendah pada stasiun C sebesar 9,2 × 102 CFU/mL. Dua isolat yaitu MTB.1a dan
MTD.1c dilakukan identifikasi molekuler menggunakan primer universal 27F dan
1492R dengan metode PCR. Hasil identifikasi menunjukkan bahwa isolat
MTB.1a merupakan Priestia megaterium dan isolat MTD.1c merupakan Bacillus
mycoides. Hasil uji kemampuan bakteri sebagai pelarut fosfat pada isolat yang
teridentifikasi dengan pengulangan sebanyak 4 kali tidak ditemukan zona bening
sehingga isolat yang ditemukan bukan termasuk golongan bakteri pelarut fosfat.

The mangrove ecosystem in Tapak is an ecosystem located along the Tapak River
in Semarang and surrounded by traditional ponds of milkfish and shrimp. The
Tapak River is a place that is suspected to be a dumping ground for industrial
waste. This location and conditions make it rich in nutrients contained in
sediments, especially in the rhizosphere, or sedimentary areas around the root.
This supports various microbial life in it, which plays a role in the process of
biogeochemical cycles and life support in mangrove ecosystems. The purpose of
this study is to isolate bacteria, determine the abundance of bacteria, and identify
the type of rhizosphere bacteria, which are then tested for their ability as
phosphate solvents. This research uses an exploratory approach. The sampling
was carried out in November 2021 and the sample analysis was carried out from
November 2021 to June 2022. Sediment samples were taken at 4 stations using
sediment cores. Sediment samples are then isolated on GSP media and nutrients
in order to obtain pure isolates. A total of 22 isolates were obtained, with 11
Gram-positive isolates and 11 Gram-negative isolates. The entire cell form
obtained was bacillus. The highest bacterial abundance was obtained at station A
at 1.44 × 104 and the lowest abundance at station C at 9.2 × 102. Two isolates,
namely MTB.1a and MTD.1c, were identified with molecular identification using
universal primers 27F and 1492R using the PCR method. The identification
results showed that the MTB.1a isolate was Priestia megaterium and the MTD.1c
isolate was Bacillus mycoides. The test results of the ability of bacteria as
phosphate solvents in isolates identified with repeated 4 times did not find clear
zones, so that the isolates found were not included in the group of phosphate
solvent bacteria.
