Abstract

RINGKASAN
Fani Alifya Nurul Ichsan. 26040118120012. Penentuan Kandungan Tanin Total
Daun Tua dan Muda Mangrove Rhizophora apiculata Blume., Avicennia alba
Blume. dan Bruguiera gymnorrizha Lamk. Secara Spektofotometri (AB Susanto
dan Sri Sedjati).
Mangrove merupakan jenis tumbuhan yang dapat tumbuh di daerah yang
ektrim, karena mangrove memiliki senyawa yang dapat melindungi diri dari
kerusakan, salah satunya adalah metabolit sekunder tanin. Tanin merupakan
senyawa bioaktif yang termasuk dalam golongan polifenol dan berperan sebagai
pertahanan terhadap mikroorganisme. Mangrove yang digunakan ada 3 spesies,
yaitu Rhizophora apiculata Blume., Avicennia alba Blume., Bruguiera
gymnorrizha Lamk. Penelitian ini bertujuan untuk mengidentifikasi kandungan
senyawa tanin yang terkandung dalam daun tua dan muda 3 spesies mangrove
tersebut.
Metode yang diterapkan pada penelitian ini adalah diskriptif kuantitatif.
Penelitian ini dilakukan terhadap 3 daun mangrove tua dan muda yang
mendominasi ekosistem mangrove Morodemak. Penentuan kandungan tanin
menggunakan metode spektofotometri dilakukan di Laboratorium Ilmu Nutrisi
Pakan, Fakultas Peternakan dan Pertanian, Universitas Diponegoro.
Spektofotometer UV-VIS merupakan alat yang biasa diguakan untuk menganalisis
bahan kimia, baik secara kuantitatif maupun kualitatif. Sebelum dilakukan analisis
kuantitatif, terlebih dahulu dilakukan penentuan panjang gelombang maksimum.
Panjang gelombang maksimum (λmax) adalah panjang gelombang pada saat
serapannya maksimum dengan cara membaca serapan larutan standar asam tanat
dan kemudian diubah-ubah panjang gelombangnya. Tanin tidak berwarna dan tidak
memiliki serapan di daerah visible, maka dari itu diberikan reagen Folin Denis agar
muncul warna dan agar dapat dibaca pada daerah visible, yaitu pada λmax 744,6
nm.
Hasil yang didapatkan yaitu Rhizophora apicullata Blume. tua didapatkan
rata-rata sebesar 2,45 mg/g, Rhizophora apicullata Blume. muda didapatkan rata�rata sebesar 34,85 mg/g, Avicennia alba Blume. tua didapatkan rata-rata sebesar
12,98 mg/g, Avicennia alba Blume. muda didapatkan hasil rata-rata sebesar 11,88
mg/g, Bruguiera gymnorrizha Lamk. tua didapatkan rata-rata sebesar 0,71 mg/g
dan Bruguiera gymnorrizha Lamk. muda didapatkan rata-rata sebesar 5,14 mg/g.
Berdasarkan hasil tersebut dapat disimpulkan bahwa kandungan nilai tanin daun
mangrove muda lebih tinggi daripada daun mangrove tua.

Content of Old and Young Mangrove Leaves Rhizophora apiculata Blume.,
Avicennia alba Blume. and Bruguiera gymnorrizha Lamk. Using Spectophotometry
UV-VIS (AB Susanto dan Sri Sedjati).
Mangroves are plant species that can grow in extreme areas, because
mangroves have compounds that can protect themselves from damage, one of which
is a secondary metabolite of tannins. Tannins are bioactive compounds that belong
to the polyphenol group and act as a defense against microorganisms. There are 3
mangroves species used, are Rhizophora apiculata Blume., Avicennia alba Blume.,
Bruguiera gymnorrizha Lamk. This study aims to identify the content of tannin
compounds contained in the old and young leaves of the 3 mangroves species.
The method applied in this research is descriptive quantitative. This research
was conducted on 3 old and young mangrove leaves that dominate the Morodemak
mangrove ecosystem. Determination of tannin content using the
spectrophotometric method was carried out at the Feed Nutrition Science
Laboratory, Faculty of Animal Husbandry and Agriculture, Diponegoro
University. UV-VIS spectrophotometer is a tool commonly used to analyze
chemicals, both quantitatively and qualitatively. Prior to quantitative analysis, the
maximum wavelength was determined. The maximum wavelength (λmax) is the
wavelength at which the absorption is maximum by reading the absorption of a
standard solution of tannic acid and then changing the wavelength. Tannins are
colorless and have no absorption in the visible region, therefore Folin Denis
reagent is given to make the color appear and so that it can be read in the visible
region, namely at max 744,6 nm.
The results obtained were Rhizophora apicullata Blume. the average age was
2.45 mg/g, Rhizophora apicullata Blume. Young adults obtained an average of
34.85 mg/g, Avicennia alba Blume. the average age was 12.98 mg/g, Avicennia
alba Blume. Young adults obtained an average yield of 11.88 mg/g, Bruguiera
gymnorrizha Lamk. the average age was 0.71 mg/g and Bruguiera gymnorrizha
Lamk. young adults obtained an average of 5.14 mg/g. Based on these results, it
can be concluded that the tannin content of young mangroves leaves is higher than
that of old mangroves leaves.
