Haydie Ilham Bagaskara. 26010216140054. Pengaruh Pemberian Ammonia pada
Sperma terhadap Motilitas dan Derajat Pembuahan Kerang darah (Anadara granosa)
(Fajar Basuki dan Diana Chilmawati).
Kerang darah (Anadara granosa) merupakan kerang yang hidup di perairan
dangkal dan substrat berlumpur, kerang darah dibudidayakan karena memiliki nilai
jual yang tinggi . Kerang darah banyak diminati oleh masyarakat karena memiliki
rasa yang lezat dan nilai jual yang ekonomis. Untuk memenuhi kebutuhan permintaan
pasar kerang darah yang berkualitas, maka penguasaan teknik budidaya khususnya
teknik pembuahan kerang darah agar menghasilkan kerang darah dengan cepat yang
berkualitas.
Penelitian ini bertujuan untuk mengetahui pengaruh amonia terhadap motilitas
sperma dan pembuahan kerang darah serta mengetahui dosis amonia terbaik terhadap
motilitas sperma dan pembuahan kerang darah. Larutan amonia dipilih karena sebagai
pembantu menambah rangsangan pemijahan kerang darah dengan meningkatkan pH
media sehingga sel sperma dapat membuahi sel telur. Adapun pelaksanaannya
dilakukan pada bulan April-Mei 2021 di Laboratorium Akuakultur Universitas
Diponegoro.
Kerang darah (Anadara granosa) diambil dari perairan laut di Wedung,
Kabupaten Demak, Jawa Tengah, kemudian dipelihara dengan pemberian pakan
berupa Chlorella vulgaris selama satu minggu. Perlakuan pemberian larutan amonia
dilakukan setelah diambil masing-masing sel sperma dan sel telur dari kerang jantan
dan betina yang dibedah. Metode penelitian digunakan ialah eksperimental
menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan yaitu perlakuan
A (tanpa pemberian amonia pada sperma), perlakuan B (pemberian amonia dosis
0,005% dari volume larutan sperma), perlakuan C (pemberian larutan amonia dosis
0,010% dari volume larutan sperma) dan perlakuan D (pemberian larutan amonia
dosis 0,015% dari volume larutan sperma) dengan 3 kali pengulangan.
Berdasarkan hasil dari penelitian menunjukan bahwa pemberian larutan amonia
berpengaruh nyata (P<0,05) terhadap motilitas sperma kerang darah namun tidak
berpengaruh terhadap pembuahan telur kerang darah, persentase motilitas tertinggi
pada perlakuan B (pemberian amonia 0,005% dari volume larutan sperma) yaitu
53.54 - 4.87. Pemberian larutan amonia ini tidak berpengaruh terhadap pembuahan
telur kerang darah hal ini diduga karena pH media setelah diberi larutan amonia tidak
dapat diterima oleh telur kerang darah untuk mengalami pembuahan.

Blood clams (Anadara granosa) are clams that live in shallow waters and
muddy substrates, blood clams are cultivated because they have a high selling
value. Blood clams are in great demand by the public because they have a
delicious taste and economical selling value. To meet the needs of the market
demand for quality blood clams, mastery of cultivation techniques, especially the
fertilization technique of blood clams in order to produce quality blood clams
quickly.
This study aims to determine the effect of ammonia on sperm motility and
fertilization of green mussels and determine the best dose of ammonia on sperm
motility and fertilization of green mussels. Ammonia solution was chosen because
as a helper it increases the stimulation of spawning blood clams by increasing the
pH of the media so that sperm cells can fertilize eggs. The implementation will be
carried out in April-May 2021 at the Aquaculture Laboratory of Diponegoro
University.
Blood clams (Anadara granosa) were taken from sea waters in Wedung,
Demak Regency, Central Java. Then they were reared by feeding with Chlorella
vulgaris for one week. The treatment of giving ammonia solution was done after
each sperm cell and egg cell were taken from dissected male and female shells.
The research method used was experimental using Completely Randomized
Design (CRD) with 4 treatments, namely treatment A (without giving ammonia to
sperm), treatment B (administration of ammonia at a dose of 0.005% of the
volume of sperm solution), treatment C (administration of ammonia solution at a
dose of 0.010% of the volume of sperm solution) and treatment D (administration
of ammonia solution at a dose of 0.015% of the volume of sperm solution) with 3
repetitions.
Based on the results of the study showed that the administration of ammonia
solution had a significant effect (P<0.005) on sperm motility of blood clams but
had no effect on fertilization of blood clams eggs. The highest percentage of
motility in treatment B (administration of ammonia 0.005% of the volume of sperm
solution) was 53.54 - 4.87 . The administration of this ammonia solution had no
effect on the fertilization of blood clam eggs. This is presumably because the pH of
the media after being given an ammonia solution was not acceptable for the blood
clam eggs to undergo fertilization.
