Ikan mas (Cyprinus carpio) merupakan salah satu spesies ikan yang hidup di air
tawar dan mempunyai peluang pengembangan budidaya besar untuk meraih potensi pasar
yang terus meningkat dan bernilai ekonomi yang tinggi. Kegiatan transportasi ikan adalah
salah satu kegiatan penting dalam penyediaan benih.Transportasi ikan yaitu menempatkan
ikan dalam lingkungan baru yang berbeda dengan lingkungan asalnya disertai perubahan
sifat lingkungan yang mendadak. Penambahan ekstrak daun jambu biji diduga mengandung
metabolit sekunder flavonoid, tanin, alkaloid, steroid dan saponin. Flavonoid (kuersetin)
yang berperan sebagai agen antimetabolit pada ikan. daun jambu biji mengandung
komponen flavonoid yaitu kuersetin. Kuersetin mengandung 5 gugus hidroksil bebas dan
merupakan senyawa polar.Daun jambu biji mengandung alkaloid, terpenoid, glikosida,
tanin, dan flavonoid yang memiliki aktivitas antidiabetes dan memiliki tinggi antioksidan.
Kandungan lainnya yaitu minyak lemak 6% dan minyak atsiri 0,4%, damar 3%, tanin 9%
dan lainnya. Daun jambu biji juga mengandung zat lain selain tanin, seperti asam ursolat,
asam psidiolat, asam kratogolat, asam oleanolat, asam guajaverin, dan vitamin.
Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun jambu
biji terhadap hemoglobin dan kelulushidupan benih dan mengetahui dosis terbaik ekstrak
daun jambu. Penelitian ini dilaksanakan pada Agustus-September 2021 di Balai Benih Ikan
(BBI) Mijen, Semarang, Jawa Tengah. Bahan uji yang digunakan adalah larutan teh dan
telur ikan nila yang berasal dari BBI Mijen, Semarang. Metode yang digunakan adalah
eksperimen dengan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan masingmasing 3 ulangan, perlakuan A (0%), B ( 0,10%), C (0,20%), D (0,30%). Data yang diamati
adalah hemoglobin darah, Kelulushidupan kualitas air, lama waktu pingsan dan lama waktu
pulih.
Hasil penelitian menunjukkan bahwa pemberian ekstrak daun jambu biji pada saat
proses transportasi tidak berpengaruh nyata terhadap perubahan nilai hemoglobin dan
kelulushidupan benih ikan mas. Nilai selisih perubahan hemoglobin pada perlakuan 0%;
0,1%; 0,2%; dan 0,3% sebesar 1,37 ± 1,48 gr/ dL; -0,87 ± 3,19 gr/ dL; 3,57 ± 1,88 gr/ dL;
dan 4,73 ± 2,57 gr/ dL serta nilai kelulushidupan sebesar 90,67 ± 2,31%; 94,00 ± 4,00%;
94,67 ± 2,31% dan; 95,33 ± 3,06%. Benih pada perlakuan 0,2% dan 0,3% mulai
menunjukkan efek lemas dan pingsan serta lambatnya pergerakan operkulum pada menit
ke-5 hingga 10. Perlakuan 0,1% menunjukkan ikan pulih pada menit ke-10. Hasil
parameter suhu 24,00

C, pH 7,00 – 8,10 dan DO 3,78-5,20 mg/L masih tergolong
optimal untuk pemeliharaan benih ikan mas.

guajava) on the Transportation Process Hemoglobin and Survival rate (Cyprinus carpio).

(Cyprinus carpio) is one of the fish species that live in fresh water and has great
opportunities for developing aquaculture to reach market potential that continues to
increase and has high economic value.Fish transportation activities are one of the
important activities in providing seeds. Fish transportation is placing fish in a new
environment that is different from their original environment accompanied by sudden
changes in environmental characteristics. The addition of guava leaf extract is thought to
contain secondary metabolites of flavonoids, tannins, alkaloids, steroids and saponins.
Flavonoid (quercetin) which acts as an antimetabolite agent in fish. Guava leaves contain
a flavonoid component, namely quercetin. Quercetin contains 5 free hydroxyl groups and
is a polar compound. Guava leaves contain alkaloids, terpenoids, glycosides, tannins, and
flavonoids which have antidiabetic activity and are high in antioxidants. Other ingredients
are 6% fatty oil and 0.4% essential oil, 3% resin, 9% tannin and others. Guava leaves also
contain other substances besides tannins, such as ursolic acid, psidiolat acid, kratogolat
acid, oleanolat acid, guajaverin acid, and vitamins.
This study aims to determine the effect of guava leaf extract on hemoglobin and
survival of seeds and determine the best dose of guava leaf extract. This research was
conducted in August-September 2021 at the Mijen Fish Seed Center (BBI), Semarang,
Central Java.The test materials used were a solution of tea and tilapia eggs from BBI
Mijen, Semarang. The method used is an experiment with Completely Randomized Design
(CRD) with 4 treatments and 3 replications each, treatment A (0%), B (0.10%), C (0.20%),
D (0.30 %). The data observed were blood hemoglobin, water quality of life, duration of
fainting and recovery time.
The results showed that the administration of guava leaf extract during the
transportation process had no significant effect on changes in the hemoglobin value and
survival rate of carp fry. The value of the difference in hemoglobin changes in treatment is
0%; 0.1%; 0.2%; and 0.3% at 1.37 ± 1.48 g/dL; -0.87 ± 3.19 g/dL; 3.57 ± 1.88 g/dL; and
4.73 ± 2.57 g/dL and a survival value of 90.67 ± 2.31%; 94.00 ± 4.00%; 94.67 ± 2.31%
and; 95.33 ± 3.06%. Seeds in the 0.2% and 0.3% treatments began to show the effects of
weakness and fainting and the slow movement of the operculum at 5 to 10 minutes. The
0.1% treatment showed the fish recovered at 10 minutes. The parameters of temperature
24,000 – 28,000 C, pH 7,00 – 8,10 and DO 3,78-5,20 mg/L are still considered optimal for
the maintenance of carp fry.
