Abstract

Gorgonian Alcyonium sp. Terhadap Patogen Penyakit Kulit (Agus Sabdono dan
Mada Triandala Sibero)
Penyakit kulit merupakan penyakit yang menginfeksi lebih dari 30%
populasi di dunia. Penyakit kulit dapat disebabkan oleh mikroflora kulit seperti
Cutibacterium acnes dan Staphylococcus epidermidis penyebab jerawat dan
diderita 85% populasi manusia di dunia. Kolonisasi khamir mikroflora kulit
seperti Malassezia furfur juga dapat menyebabkan ketombe yang diderita 30%
populasi manusia di dunia. Khamir mikroflora lain, Candida albicans penyebab
kandidiasis diderita oleh 40% populasi manusia di dunia. Penggunaan antibiotik
merupakan salah satu upaya penanganan penyakit kulit. Namun, permasalahan
yang terjadi adalah timbulnya resistensi bakteri dan jamur patogen terhadap
antibiotik yang umum digunakan seperti ketoconazole, fluconazole, tetracyclines,
macrolides, trimethoprim/sulfamethoxazole, dan clindamycin. Oleh karena itu,
dibutuhkan kajian senyawa antibiotik baru untuk melawan patogen tersebut. Salah
satu sumber antibiotik baru yang menarik untuk dikaji adalah bakteri asosiasi
gorgonian Alcyonium sp. Bakteri asosiasi gorgonian Alcyonium sp. diduga mampu
menghasilkan senyawa antimikroba.
Penelitian ini berfokus untuk mengisolasi bakteri asosiasi gorgonian yang
memiliki kemampuan melawan Cutibacterium acnes, Staphylococcus
epidermidis, Malassezia furfur dan Candida albicans serta melakukan pendekatan
secara molekuler. Metode yang digunakan dalam penelitian ini yakni deskriptif
eksploratif laboratory. Prosedur penelitian ini meliputi pengambilan sampel,
isolasi dan purifikasi bakteri, uji antimikroba metode agar plug, karakterisasi
morfologi, serta identifikasi molekuler. Hasil dari penelitian menunjukkan bahwa
terdapat delapan isolat yang didapat dari Gorgonian Alcyonium sp. dari perairan
Pulau Seruni, Karimunjawa. Satu isolat bakteri asosiasi Gorgonian Alcyonium sp.
memiliki aktivitas antimikroba melawan Cutibacterium acnes, Staphylococcus
epidermidis, serta Candida albicans. Hasil identifikasi molekuler menunjukkan
bahwa isolat SE.4.2 memiliki kemiripan sebesar 98,96% dengan
Pseudoalteromonas shioyasakiensis.

Bacteria from Gorgonian Alcyonium sp. Against Skin Diseases (Agus Sabdono
dan Mada Triandala Sibero)
Skin disease affects more than 30% populations in the world. Skin diseases
can be caused by skin microflora such as Cutibacterium acnes and Staphylococcus
epidermidis which cause acne and affects 85% of the world's population. Yeast
colonization of skin microflora such as Malassezia furfur can also cause dandruff
which affects one third of the world's population. Other yeast microflora, Candida
albicans causes candidiasis suffered by 40% of the world's population. One of the
common treatment for skin diseases is the application of antibiotic products.
However, the problem that occurs is the emergence of resistance of pathogenic
bacteria and fungi to commonly used antibiotics such as ketoconazole, fluconazole,
tetracyclines, macrolides, trimethoprim/sulfamethoxazole, and clindamycin.
Therefore, it is necessary to study new antibiotic compounds to fight these
pathogens. One of the interesting new sources of antibiotics to study is the
gorgonian-associated bacteria Alcyonium sp. Gorgonian-associated bacteria
Alcyonium sp. are known to be capable of producing antimicrobial compounds.
This research focuses on isolating gorgonian-associated bacteria that have
the ability produce antimicrobial compounds against Cutibacterium acnes,
Staphylococcus epidermidis, Malassezia furfur and Candida albicans and also
carry out a molecular approach. The method used in this research is descriptive
exploratory laboratory. The procedure of this research includes sampling,
isolation and purification of bacteria, antimicrobial test with agar plug method,
morphological characterization, and molecular identification. The results of the
study showed that there were eight isolates obtained from the gorgonian
Alcyonium sp. from the waters of Seruni Island, Karimunjawa. One isolate of
gorgonian associated-bacteria Alcyonium sp. has antimicrobial activity against
Cutibacterium acnes, Staphylococcus epidermidis, and Candida albicans. The
results of molecular identification showed that isolate SE.4.2 had a similarity of
98.96% with Pseudoalteromonas shioyasakiensis.
