Achmad Baharudin. 26010118140067. Isolasi dan Identifikasi Bakteri Toleran
Logam Berat Pb (Timbal) dan Cu (Tembaga) dari Sedimen Mangrove di Mangrove
Tapak, Semarang. (Niniek Widyorini dan Diah Ayuningrum)
Ekosistem mangrove Tapak, Semarang merupakan ekosistem rehabilitasi yang
sebelumnya merupakan lahan tambak yang dijadikan Kawasan ekowisata.
Ekosistem mangrove memiliki fungsi ekologi yaitu penyerap karbon, nitrogen dan
fosfor; menyaring dan mengurangi nutrisi terlarut; serta memerangkap logam berat
dalam sedimen mangrove. Ekosistem mangrove dapat mengalami pencemaran
logam berat yang berasal dari bahan industri dan hasil pembakaran bahan bakar
kendaraan. Logam berat yang ditemukan pada daerah ekosistem mangrove
sebagian besar merupakan logam timbal (Pb) dan tembaga (Cu). Tujuan penelitian
ini adalah untuk mengetahui jumlah isolat yang didapatkan dari sedimen mangrove
Tapak, mengetahui isolat bakteri yang toleran dengan logam berat, dan mengetahui
bakteri spesies bakteri yang toleran dengan logam berat. Penelitian ini dilakukan
pada bulan Desember 2021 - Juli 2022. Metode yang digunakan yaitu metode
deskriptif dan metode pengambilan sampel menggunakan metode purpose
sampling dengan mengambil beberapa sampel sedimen. Metode isolasi dan dan
purifikasi menggunakan pengenceran bertingkat; skrining bakteri toleran logam
berat dengan media peptone yeast agar yang ditambahkan garam logam berat Pb
dan Cu; serta identifikasi molekuler bakteri dengan gen 162 rRNA yang dilakukan
di Tropical Marine Biotecnology Gedung J FPIK UNDIP. Hasil penelitian
didapatkan hasil isolasi sebanyak 18 isolat bakteri asal sedimen mangrove, 2 isolat
bakteri yang memiliki toleran paling tinggi terhadap logam berat dengan kode A1-B dan C1-A. Kesimpulan yang dapat diambil dari hasil penelitain ini yaitu bahwa
bakteri pada sedimen mangrove Tapak, Semarang dapat hidup pada konsetrasi
logam Pb 70-560 ppm dan pada kosentrasi logam Cu 30-120.

at Mangrove Tapak, Semarang. (Niniek Widyorini dan Diah Ayuningrum)
The Tapak mangrove ecosystem, Semarang is a rehabilitation ecosystem that was
previously a pond area that was used as an ecotourism area. Mangrove ecosystems
have ecological functions, namely carbon, nitrogen and phosphorus absorption;
filter and reduce dissolved nutrients; and trapping heavy metals in mangrove
sediments. Mangrove ecosystems can experience heavy metal pollution from
industrial materials and the results of burning vehicle fuels. Heavy metals found in
mangrove ecosystem areas are mostly lead (Pb) and copper (Cu). The purpose of
this study was to determine the number of isolates obtained from the mangrove
sediments of Tapak, to determine the bacterial isolates tolerant to heavy metals,
and to determine the bacterial species tolerant to heavy metals. This research was
conducted in December 2021 - July 2022. The method used is descriptive method
and the sampling method uses the purpose sampling method by taking several
sediment samples. Isolation and purification methods use graded dilutions;
screening of heavy metal tolerant bacteria with peptone yeast agar media with
added heavy metal salts of Pb and Cu; and molecular identification of bacteria with
the 162 rRNA gene carried out at Tropical Marine Biotecnology Building J FPIK
UNDIP. The results showed that 18 isolates of bacteria from mangrove sediments
were isolated, 2 isolates of bacteria that had the highest tolerance for heavy metals
with codes A1-B and C1-A. The conclusion that can be drawn from the results of
this research is that bacteria in the mangrove sediments of Tapak, Semarang can
live at a metal concentration of Pb 70-560 ppm and at a metal concentration of Cu
30-120.
